Scientists 'delete' HIV virus from human DNA for the first time

It inserts its deadly genome permanently into its victims' DNA, forcing them to require medical treatment for the rest of their life.
But now, for the first time, researchers in Philadelphia have found a way to completely delete HIV from human cells by ‘snipping’ them out.

The team of Temple University School of Medicine said the breakthrough marks the first successful attempt to eliminate latent HIV-1 virus from human cells – and could be a cure for other latent infections.





HOW THE PROCESS WORKS

Researchers based the two-part HIV-1 editor on a system that evolved as a bacterial defence mechanism to protect against infection.

When deployed, a combination of a DNA-snipping enzyme called a nuclease and a targeting strand of RNA called a guide RNA (gRNA) hunt down the viral genome and remove the HIV-1 DNA.

Dr Khalili's lab engineered a 20-nucleotide strand of gRNA to target the HIV-1 DNA and paired it with a DNA-sniping enzyme called Cas9 and used to edit the human genome.

From there, the cell's gene repair machinery takes over, soldering the loose ends of the genome back together – resulting in virus-free cells.

When deployed, a combination of a DNA-snipping enzyme called a nuclease and a targeting strand of RNA called a guide RNA (gRNA) hunt down the viral genome and remove the HIV-1 DNA.

From there, the cell's gene repair machinery takes over, soldering the loose ends of the genome back together – resulting in virus-free cells.

'Since HIV-1 is never cleared by the immune system, removal of the virus is required in order to cure the disease,' explained Dr Khalili.

These molecular tools also hold promise as a therapeutic vaccine; cells armed with the nuclease-RNA combination proved impervious to HIV infection.

Worldwide, more than 33 million people have HIV, including more than 1 million in the United States.

Every year, another 50,000 Americans contract the virus, according to the U.S. Centers for Disease Control and Prevention.

In the UK, around 100,000 people were living with HIV in the UK in 2013. That’s around one person in 665.

Although highly active antiretroviral therapy (HAART) has controlled HIV-1 for infected people in the developed world over the last 15 years, the virus can rage again with any interruption in treatment.

I became very close to him about 5 years ago when he invited me to run his election campaign as director general which he won and also the reelection too. All through this time I saw in him a gentleman per excellence, incisive, critical, detailed, compassionate, supportive, kind, a family man and detribalized Nigerian. Astute orator and an encyclopedia of knowledge and information. 

This man who turns 50 today is a talented writer, producer, award-winning journalist and president of Association of Movie Producers of Nigeria Zik Zulu Okafor. As you celebrate your this joyful day with your charming wife Adora and 3 lovely kids, I wish you many more years of blessings and successes.

Sean Connery, the original James Bond, turns 85

Scottish actor Sir Sean Connery, who played the first ever James Bond in the super hit series, turned 85 on 26 August. He was named the sexiest man alive in 1989 and 10 years later, was voted as the sexiest man of the century.
The octogenarian was extremely popular in his early days, especially with women, following the success of his first Bond movie titled Dr No, which released in 1962. Off camera, the actor has managed to flaunt his style and elegance even through his golden years. Connery is one of those rare actors, who can take his shirt off and still leave a good impression at every age.

Connery, has received some of Hollywood's biggest honours including an Oscar for his supporting role as Jimmy Malone in the mafia drama The Untouchables. He has also won three Golden Globe and two BAFTA awards.

Today, we will start a new sub-series under the Pursuit of Purpose. It is: How to Deal with Discouragement and Self Doubt.

In the pursuit of purpose, there is a place for a mentor, an encourager, a compass or navigator to help you stay on track and build in you a compelling passion to win. You must, however, be careful in your choice of a mentor.

If you make a failure your mentor, you are guaranteed to multiply his failure. It is a sure way to become a Mega Failure! You, therefore, must carefully choose a success as a mentor. You may also choose a Mega Success as your mentor.

This can affect you in either of two ways; you can either get so fired up to want to do everything possible to match or exceed him. On the other hand, you may be overwhelmed by his great accomplishments such that you just live in his shadows all your days and never actualize your own potentials

The Bible recognizes the place of mentors. That is why we are enjoined to look onto Jesus. He is our ultimate role model. It is okay to have human role models, but the most profound examples for successful living and purpose fulfillment can only be gotten from Jesus.

Make Him your role model and find yourself a human role model prayerfully. It will help in your pursuit

SERENA EDGES HALEP & WINS CINCINNATI

Serena Williams held off a fierce challenge from Simona Halep to win her fifth WTA title of the year, the milestone 69th WTA title of her career, keep an amazing finals streak going and head into the US Open - where she's going for her 22nd major - brimming with confidence.
There were scratchy moments during the week, including a big quarterfinal scare against Ana Ivanovic, but when it came down to the final Williams brought out her very best tennis, coming back from 3-1 down to take the first set and then surviving a marathon 67-minute second set to prevail, 6-3, 7-6(5).
The No.3-seeded Halep was on her game, her 18 winners outweighing 14 unforced errors, but the No.1-seeded Williams finished with an incredible +17 winners-to-unforced errors differential, 44 to 27.



Halep didn't leave empty-handed - by reaching the final, she'll now return to World No.2 on the new WTA Rankings, which determine the US Open seeds - meaning she can't play Williams until the final.
And she had some words of encouragement as the World No.1 embarks on her historic quest.
"I have to say congratulations to the champion, Serena, and I wish you good luck at the US Open," the always-classy Romanian said. "I know that you can win all four - so good luck, and be strong."
Williams was just as complimentary in her speech. "Thank you Simona for playing such a good match today, and always being such an incredible opponent and incredible sportswoman," she said. "I'm always honored to play against you and always have such a wonderful match whenever we play.
"I've also got to say, I don't get support everywhere I play, but I get so much support here in Cincinnati that it makes me want to come back forever and ever, so thank you to all the fans today!"
Williams now has a WTA-leading five titles on the season, having already won the Australian Open, Miami, the French Open and Wimbledon, and she's now won 69 WTA titles in her career, which sets her apart from Evonne Goolagong as standalone fifth place for most WTA titles in the Open Era, trailing only Martina Navratilova (167), Chris Evert (154), Steffi Graf (107) and Margaret Court (92).
She's also won 15 finals in a row now - her last loss in a final came right here in Cincinnati two years ago to Victoria Azarenka, and by the slimmest of margins - a 2-6, 6-2, 7-6(6) final scoreline.

Malala Yousafzai, Shot By Taliban For Supporting Girls' Education, Celebrates Straight-A's


The youngest-ever winner of the Nobel Peace Prize has turned down hundreds of interviews to focus on schoolwork.

Teenage Nobel Peace Prize winner Malala Yousafzai received straight-A’s in her General Certificate of Secondary Education (GCSE) examinations, the Daily Pakistan reports. The GCSE’s are standardized subject tests taken by secondary school students in England, Wales and Northern Ireland. The scores influence a test-taker’s future educational pursuits or professional prospects.
Yousafzai’s father, Ziauddin Yousafzai, tweeted his daughter’s grades shortly after they were released Thursday:
Passing grades range from A* (the highest grade, similar to an American A-plus) to “F,” while a failing mark is a “U.”
The Pakistani 18-year-old attends a private girls’ school in Birmingham, England. Her family has lived in the city since Yousafzai was transferred to a Birmingham hospital following a 2012 attack by a Taliban gunman in her home country. When Yousafzai -- who had been an outspoken advocate for girls’ education for several years -- was riding a bus home from school, a group of men boarded the vehicle and asked for her specifically. One man then shot her in the head.
After her recovery, she started the Malala Fund, a nonprofit devoted to girls’ education worldwide. She also won the Nobel Peace Prize in 2014, becoming the youngest Nobel laureate in history.
On top of her other achievements, she’s managed to prioritize schoolwork, turning down hundreds of interviews and speaking engagements in order to study, according to The Guardian.
The Pakistan-based Express Tribune wrote that Yousafzai has “made the country proud once again.”

The 28-year-old has accumulated three Champions Leagues and five La Liga titles, as well as being a world and European champion on the international stage.

Born in Tenerife, the diminutive Pedro, whose full name is Pedro Eliezer Rodríguez Ledesma, signed for BarÏ‚a’s academy in 2004 and made his first-team debut in January 2008 after catching the eye for their B team.          

By 2009/10, the two-footed, fast and direct winger had become an important part of the Barcelona team. He was the first player ever to score in the Spanish league, Copa del Rey, Champions League, both the European and Spanish Super Cups as well as the World Club Cup.

His brilliant performances at club level saw him secure a place in the Spain squad for the 2010 World Cup in South Africa. Pedro started both the semi-final and final, and was a key figure in their victory over the Netherlands.


A year later he scored the opening goal in the Champions League final at Wembley as Barςa beat Manchester United 3-1.

He was a substitute alongside then Chelsea players Fernando Torres and Juan Mata in Spain’s Euro 2012 victory over Italy. The winger was also part of the squad which travelled to Brazil for the 2014 World Cup.


Pedro came on as a late substitute in Barcelona’s most recent major success - May’s Champions League final win over Juventus in Berlin. He concluded his time at the Catalan club in the most memorable manner, scoring the winning goal in their 5-4 UEFA Super Cup triumph over Sevilla.

NNPC to become sole importer of fuel ”Presidency"

The Central Bank of Nigeria (CBN) Governor, Mr. Godwin Emefiele, said the government was doing everything to ensure that the Nigerian National Petroleum Corporation (NNPC) become sole importer of petroleum products.
The CBN Chief  said: “The President came on board and said that we will work very hard to reduce importation of petroleum products by ensuring that our refineries work. Our refin¬eries are working now.

“The Warri and Port Harcourt refineries have started producing. They have not obtained the optimal capacity but they will. The Kaduna refinery will start working this month.
“Now, there are other actions that the Presidency is putting in place to ensure that we reduce importation of petroleum products where the NNPC will solely, almost solely be responsible for procuring refined petroleum.
 ”Those who are importing petroleum products will only just need to go to the NNPC and pick up petroleum products.
“So, in that area, I would say that we are already moving in the direction of reducing the import of pe¬troleum products. And we will achieve it.”


North Korea's Vice Premier Choe Yong Gon Executed



North Korea's vice premier was executed by firing squad this year after showing discontent with the policies of the country's leader Kim Jong Un, a South Korean media report said on Wednesday.
Yonhap News Agency cited an unnamed source as saying that the 63-year-old Choe Yong Gon, a former delegate for North-South cooperation, was executed, marking another death of a senior official in a series of high-level purges since Kim Jong Un took charge in late 2011.
The Yonhap report said Choe had expressed disagreement with Kim's forestry policies in May and had shown poor work performance. It provided no further details.
South Korea's Unification Ministry, which handles the country's ties with North Korea, said in a text message received by Reuters that Choe had not been spotted in public for about eight months, and that it was closely monitoring the situation.
South Korea's National Intelligence Service declined to comment on the report to Reuters.
The South Korean spy agency told lawmakers in May that North Korea had executed its defense chief by putting him in front of an anti-aircraft gun at a firing range.
Choe was appointed vice-premier last year, North Korea's state-run KCNA news agency reported previously.
Yonhap said the source also said the reclusive state had publicly executed a senior Workers' Party official in September.
Choe had worked on inter-Korean affairs in 2000s, leading the North's delegation in joint economic cooperation committees with South Korea between 2003 and 2005.
He attended the 2004 opening ceremony of the Kaesong Industrial Complex, a factory park jointly run with Seoul that is the last remaining joint project of the two countries.

(Hopefully, it should be on Nollywood Movies on B SKYB in a few months for those in London and Europe and will hit the Comcast bouquet in America soon enough too)

“And if a man also strive for masteries, yet is he not crowned, except he strive lawfully.”
Today, we will look at the last factor that triggers the desire for Short Cuts. This is what I call Scarcity Mentality! This is a major mental barrier that the world especially Africans need to recover from. This is the belief that that the benefits or the resources are not enough and so cannot go round.
It then becomes the survival of the fastest and the fiercest! I need to get in there faster than everybody else and grab as much as I can since I know it would never be enough. In this madness, we forget that once we take more than our fair share, we deprive other people their legitimate portion.
Unfortunately, people get celebrated for being the fastest and the fiercest thieves. We tend to blame and punish those who are playing strictly by the rules. We call them names. We make them feel like they have committed the abominable! Our society has suffered a travesty of values. We celebrate the rogues and vilify the right.
That is why civil servants take short cuts; private sector professionals take short cuts; students do, lecturers are actively engaged in it; business men and women take even shorter cuts and the list goes on. No matter how much you have, a scarcity mentality tells you it is not enough and so contentment is a mirage. You must keep stealing and running until you die miserably!
If your pursuit must be lawful so you can be deemed right by heaven to obtain the prize, deliver yourself today from this worst form of poverty and you have delivered your future from stillbirth!
